  1. Antigen choice: Prioritize HA head/stem and NA for neutralization; NP/M1 for serology; M2e for universal vaccine; NS1 for immune evasion; PB1/PB2/PA for replication.
  2. Subtype/lineage: Match to A/H1N1, A/H3N2, B/Victoria, B/Yamagata; use conserved HA stem, M2e for cross-protection studies.
  3. Expression: Mammalian/insect for HA/NA to retain glycosylation/trimer; bacterial/mammalian for NP, M1, polymerase with activity validation.
