Nipah virus (NiV) and Hendra virus (HeV) are enveloped, non-segmented, negative-sense RNA paramyxoviruses of the genus Henipavirus (~18.2 kb genome). NiV was first identified in 1999 following a large encephalitis outbreak in Malaysia/Singapore (case fatality 40–75%), while HeV was identified in 1994 in Australia (case fatality ~57%). Both are bat-borne BSL-4 pathogens on the WHO R&D Blueprint priority pathogen list. Henipaviruses encode six structural proteins: the attachment glycoprotein G (binds ephrin-B2/B3 receptors), fusion protein F (class I fusogen), matrix M, nucleoprotein N, phosphoprotein P (plus accessory V, W, C proteins from the P gene), and RNA polymerase L. The G protein is the primary target for neutralizing antibodies and vaccines — Nipah G is the immunogen in the leading NiV vaccine candidates currently in clinical trials.

The G protein is the receptor-binding protein that engages ephrin-B2 (widely expressed) and ephrin-B3 (predominantly neural) on host cells. G is the dominant neutralizing antigen and the immunogen in all leading NiV vaccine candidates (Oxford/Janssen ChAdOx1-NiV, PHV02/mRNA, IAVI subunit). Which antibodies cross-react with both NiV and HeV?
Most of our anti-G and anti-F antibodies are cross-reactive by design — NiV and HeV G proteins share ~83% amino acid identity, and F proteins share ~89%. Key cross-reactive clones: m102.3 and m102.4 (anti-G), and mAb66 and 5B3 (anti-F). For species-specific detection, use monoclonal antibodies paired with NiV-specific or HeV-specific recombinant G/F proteins.
Which antibody combination provides the broadest protection?
Combining anti-G (blocks attachment) + anti-F (blocks fusion) provides complementary neutralization. A two-antibody cocktail of m102.3 (anti-G) + mAb66 (anti-F) covers both entry steps and is protective in animal models. Adding prefusion-specific antibodies (4H3, 1H8) may further enhance potency.
Can I work with these reagents at BSL-2?
Yes. All products are recombinant proteins and antibodies — no live or inactivated virus. Standard BSL-1/BSL-2 conditions are sufficient. Live NiV and HeV require BSL-4.
